Avian Tuberculin Observe™ 25,000 IU per ml  is approved for use in  New Zealand for the diagnosis of tuberculosis in animals by intradermal test, i.e. diagnosis of tuberculosis in fowls and pigs; comparative tuberculin test in cattle to distinguish between avian and mammalian tuberculosis infections; or as an aid in the diagnosis of Johnes’s disease.

Dosage and Administration

  • Inject intradermally. Fowls. 0.05 to 0.1mL into the wattle. Swine. 0.1mL into the base of the ear. Cattle. 0.1mL into the caudal fold as for ordinary tuberculin test.
  • For comparative test, inject simultaneously 0.1mL of Tuberculin PPD (Bovine) 1mg intradermally into the other caudal fold. The comparative test may also be carried out by injecting 0.1mL of each of the 2 tuberculins into the skin of the neck in comparable sites about 10 cm apart. Before injecting measure and record the skin thickness at the sites of injection with callipers graduated in mm. Test interpretation. In cattle and swine read the test 72 hours after injection; in fowls 48 hours after injection.
  • A positive reaction is indicated by an increase in the skin thickness, characterised by diffuse swelling.
  • In cattle a palpable diffuse swelling of the caudal fold or a calliper measured increase of 4mm or more in the skin of the neck can be regarded as positive.
  • When the comparative test is carried out and the greater reaction is produced by tuberculin PPD (Bovine), the disease is assumed to be of mammalian origin. However, a greater reaction at the site of injection of Tuberculin PPD (Avian) indicates that infection is due to either avian tuberculosis of Johnes; disease, depending on clinical signs.
  • In swine an increase to twice that of the original skin thickness is regarded as positive.
  • In fowls a soft oedematous swelling of the wattle indicates a positive reaction. The test is not reliable for testing individual birds and is suitable only as a flock test. Retesting. Do not retest in less than 60 days.
